I. Background
淋巴细胞活化基因 3 (LAG-3)，又名 CD223，是一种 I 型跨膜蛋白，属于免疫球蛋白超家族 (IgSF)。LAG-3 蛋白与 CD4 有同源结构，两者的胞外区均具有四个 Ig 样结构域，但 LAG-3 在膜远端 Ig 样结构域内还含有一种额外的环状结构，使其与MHC II的亲和力较CD4高100倍。LAG-3 在活化的 T 细胞、B 细胞、浆细胞样树突状细胞 (DC) 和 NK 细胞上表达。LAG3 可结合 MHC II类 (MHC II)分子外，也可结合纤维蛋白原样蛋白 1（FGL-1）、α-突触核蛋白原纤维 (α-syn) 和凝集素半乳糖凝集素-3 (Gal-3) 以及淋巴结窦内皮细胞 C 型凝集素 (LSECtin) 。 LAG3 可能是比 CTLA-4 或 PD-1 更好的免疫检查点抑制剂靶点，因为这两个检查点的抗体仅激活效应 T 细胞，而不会抑制 Treg 活性，而拮抗剂 LAG3 抗体既可以激活效应 T 细胞（通过下调 LAG3 抑制信号），又可以抑制诱导的（即抗原特异性）Treg 抑制活性。

II. Description
