| I. Background | |
| IL-21 是 γc 家族细胞因子，主要由滤泡辅助性 T 细胞（Tfh）等 CD4 + T 亚群产生，通 过广泛表达于 B 细胞、CD8+ T 细胞、NK 细胞的受体发挥作用。它在体液免疫中驱动生发 中心形成与高亲和力抗体产生，在细胞免疫中维持 CD8+ T 细胞功能、防止耗竭，同时增 强 NK 细胞活性，是连接先天与适应性免疫的关键枢纽。其功能呈双刃剑效应：过度激活 导致自身免疫病，信号不足则与慢性感染及肿瘤免疫逃逸相关。 IL-21 先与特异性 IL-21Rα 链结合，再募集共有 γ 链形成受体复合物，偶联的 JAK1 与 JAK3 被激活，使受体胞内段磷酸化并招募 STAT3。STAT3 磷酸化后形成同源二聚体入核， 调控 PRDM1、BCL6 等靶基因转录，介导 B 细胞浆细胞分化与 T 细胞干性维持。同时可辅 助激活 PI3K–AKT 等通路，信号强度受 SOCS 家族等负反馈精密调控。 |
