| I. Background | |
| DLK1，也被称为 Pref-1（脂肪前体细胞因子 1，preadipocyte factor l）、FA1、pG2、 ZOG（肾上腺皮质小球带特异因子，ozan glomeurlosa-specific factor），是表皮生长因子 （EGF）家族的成员之一。在胚胎发育时期广泛表达，而在出生后只表达于少数的腺体和 组织。DLK1 在细胞分化的过程中起着重要的作用，包括干细胞、造血细胞、肾上腺细胞 及神经内分泌细胞的分化。DLK1 参与调节骨生成和脂肪形成、细胞生长和肿瘤形成、创 伤愈合及修复、外周及中枢神经系统的分化，致使低分化的肿瘤进一步恶变。DLK1 可以 参与 Notch 信号通路及 MAPK 信号通路。Notch 信号通路参与细胞正常形态形成的多个过 程，包括多能祖细胞的分化、细胞凋亡、细胞增殖及细胞边界的形成，而参与肿瘤的形成。 MAPK 信号通路更是错综复杂，参与多种疾病发病及肿瘤增殖的过程。DLK1 在多种恶性 肿瘤中呈现一定频率的异常表达，包括白血病、神经母细胞瘤、神经胶质瘤、乳腺癌、结 肠癌、胰腺癌、和小细胞肺癌等。 | |
