TP53 基因编码 p53 蛋白,是调控细胞周期、DNA 修复及凋亡的核心肿瘤抑制因子。 R175H 突变是 TP53 第 175 位氨基酸由精氨酸(R)突变为组氨酸(H)的错义突变,位于其 DNA 结合域的关键功能区。该突变导致 p53 蛋白构象改变,丧失与 DNA 结合的能力, 无法正常启动细胞周期阻滞或凋亡程序,从而促进肿瘤细胞异常增殖、转移及化疗耐药。 |
