| VISTA 基因（V-set and immunoglobulin domain containing 4）是免疫调节领域的关键分 子，属于 B7 家族，与 PD-1、CTLA-4 等经典免疫检查点分子具有同源性。其编码的 VISTA 蛋白是一种 I 型跨膜蛋白，包含免疫球蛋白 V 结构域（IgV）、茎状区域、跨膜区 域和胞质结构域，其中胞外结构域（ECD）的 C-C′环富含组氨酸残基，形成 pH 依赖性结 合位点，胞质端则包含 SH3 和 SH2 结合基序及磷酸化位点，提示其信号传递功能。 在肿瘤免疫中，VISTA 高表达于骨髓来源抑制细胞（MDSCs）和肿瘤相关巨噬细胞 （TAMs），且在低氧环境下上调，通过抑制 T 细胞活性和促进免疫抑制细胞群积累，帮 助肿瘤逃避免疫监视。研究表明，VISTA 阻断可显著增强抗肿瘤免疫力，导致抗原特异性 T 细胞扩增和免疫耐受降低。 | |
