| I. Background | |

LGR5 基因（富含亮氨酸重复序列的 G 蛋白偶联受体 5）是 GPCR 家族成员，位于染 色体 12q22-23，编码 907 个氨基酸的跨膜蛋白。其胞外区含 17 个亮氨酸重复序列，通过结 合 R-spondin 增强 Wnt/β-catenin 信号通路，调控干细胞增殖、分化及肿瘤发生。
LGR5 作为 Wnt 受体的协同因子，通过与 Frizzled 和 LRP5/6 受体结合，促进[Wnt/β- catenin 信号通路激活。在正常组织中，LGR5 表达局限于肠道隐窝、毛囊等干细胞区，参 与组织再生修复；在肿瘤中，其高表达与结直肠癌、肝癌、乳腺癌等密切相关，作为癌症 干细胞标志物，促进肿瘤侵袭、转移及化疗耐药。机制上，LGR5 通过激活 Wnt 通路介导 上皮-间质转化（EMT），并抑制 p53 稳定性增强耐药性。 |
