| I. Background | |
| GHSR1a,全称为生长激素促分泌素受体1a型,是功能完整的7次跨膜受体的G蛋白 偶联受体,负责所有已知的胃饥饿素生物学效应,包括刺激食欲、促进生长激素释放、调 节能量平衡和糖脂代谢等。GHSR1a以内源性激素胃饥饿素为主要配体。它通过偶联Gq/11 蛋白,核心信号通路为PLC-β,产生IP3并动员细胞内钙离子,进而触发下游级联反应; 此外,它还能在不同细胞环境中与Gi/o或Gs等通路交互,形成一个复杂的信号转导网 络。该受体广泛分布于下丘脑、海马体、胰腺及心脏等多组织,其信号精确调控着生长激 素释放、食欲、能量代谢及神经保护等多种关键生理功能,从而成为治疗恶病质、肥胖及 相关代谢与神经疾病的重要药物靶点。 | |
