| ASCT2(又称SLC1A5)是溶质载体家族1A中的一种钠依赖性中性氨基酸反向转运体。它的核心功能是以谷氨酰胺为首选底物,与细胞内其他中性氨基酸(如丝氨酸、苏氨酸)进行交换,以此调节氨基酸稳态。 ASCT2之所以成为癌症研究的热门靶点,是因为它在多种肿瘤细胞中显著高表达,为癌细胞快速增殖所必需的谷氨酰胺代谢提供原料。其结构与功能机制复杂,最新研究发现其转运循环中可结合三个钠离子,且由于在关键跨膜结构域上缺乏变构能力,使其被限定在交换模式而非主动积累模式。 | |||
