| I. Background | |
| LIGHT 基因(TNFSF14)是肿瘤坏死因子超家族成员,编码 240 个氨基酸的Ⅱ型跨膜 糖蛋白,含 N 端胞内区、跨膜区和 C 端胞外区,可被切割为分泌型 sLIGHT。其受体包括 HVEM、LTβR 及 DcR3,通过 TRAF 介导信号通路,参与免疫调节与疾病发生。 功能上,LIGHT 刺激 T 淋巴细胞增殖、诱导树突状细胞成熟,促进 Th1 型细胞因子分 泌及 NK 细胞激活,同时可诱导肿瘤细胞凋亡。在病理过程中,LIGHT 与慢性肝病(如肝 炎、肝硬化、肝癌)密切相关:慢性肝炎患者血浆及肝组织中 LIGHT 水平升高,通过 NF- κB/TLR3 通路加剧炎症;在肝癌中,LIGHT 通过 LTβR 介导的信号促进肿瘤进展。此外, LIGHT 作为免疫佐剂可增强疫苗应答,其单克隆抗体在肿瘤免疫治疗中展现潜力。 |
