| I. Background | |
| SLC39A6 也称为 LIV-1 或 ZIP6，是锌转运蛋白家族的成员，最初被确定为乳腺癌中的 雌激素诱导基因。SLC39A6 的主要功能是将锌离子从细胞外转移至细胞内，并且是酶、转 录因子和信号分子发挥作用机制中的关键蛋白。该基因在多种肿瘤细胞（如乳腺癌、前列 腺癌、食管癌等）中高表达，而在正常组织中表达有限，成为抗体偶联药物（ADC）治疗 的潜在靶点。研究表明 SLC39A6 通过调节锌离子水平影响细胞信号传导通路（如 JAK/STAT、PI3K/AKT 等），促进肿瘤增殖、侵袭和迁移，并与上皮间质转化（EMT）相 关，通过调控 Snail 等转录因子增强细胞迁移能力。在临床试验中，靶向 SLC39A6 的 ADC 药物已显示出对转移性三阴性乳腺癌等癌症的治疗效果，凸显其在癌症治疗中的重要性。 | |
