| I. Background | |
| C-Met 属于受体酪氨酸激酶家族,是一种异二聚体,由高度糖基化的细胞外 50 kDa α 链和跨膜 140 kDa β链组成,通过二硫键连接在一起。作为一种天然的 HGF 受体,它表达 于各种上皮细胞的表面,并促进肿瘤的发生。与 HGF 结合后,两个亚基二聚化,导致两个 酪氨酸残基(Tyr1234 和 Tyr1235 )自磷酸化。酪氨酸残基的自磷酸化导致一个激活的多功 能停靠位点,该位点由 SH2 结构域、PTB 结构域和 Met 结合结构域 (MDB) 组成,该结 构域募集接头分子。 | |
