| RARα(Retinoic Acid Receptor Alpha,视黄酸受体α)是核受体超家族成员,其结构包 含高度保守的 DNA 结合域(DBD,含锌指模体) 和 配体结合域(LBD,含配体依赖性转 录激活区 AF-2),以异源二聚体形式(与 RXR 结合) 特异性识别靶基因启动子上的视黄 酸反应元件(RARE);当内源性配体全反式视黄酸(ATRA) 结合 LBD 后,触发构象变 化,释放共抑制复合物(如 N-CoR/SMRT),招募共激活因子(如 CBP/p300),进而调控胚胎发育(神经嵴分化、肢体形成)、细胞增殖/分化(如骨髓造血干细胞定向)及凋亡等 关键通路。 | |
