| I. Background | |
| CRF1R 属于 B 类 GPCR 家族,在中枢神经系统(下丘脑、杏仁核、前额叶皮层)及外 周组织(胃肠、皮肤)表达,是机体应激应答的核心调控者。CRF1R 的内源性配体促肾上 腺皮质激素释放因子(CRF)和 尿皮质素 1(UCN1)结合后,触发受体构象变化,优先偶 联 Gs 蛋白,激活腺苷酸环化酶(AC),升高 cAMP,启动 PKA/CREB 信号轴,驱动下丘 脑-垂体-肾上腺轴(HPA 轴)激活,最终促进 糖皮质激素(皮质醇)释放以应对生理/心理 压力;同时通过β-arrestin 通路调节受体脱敏内化。该受体过度激活与焦虑症、抑郁症、肠 易激综合征(IBS)及药物成瘾密切相关。 | |
