| I. Background | |
| CRF2R 属于 B 类 GPCR 超家族,在中枢(下丘脑室旁核、杏仁核、中缝背核)及外周 (心血管、骨骼肌、胃肠道)广泛分布,是应激稳态的“缓冲器”。CRFR2A 是 CRF2R 的 主要中枢亚型,由 CRHR2 基因通过外显子 1 起始转录生成,其胞外 N 端结构域(34 个氨 基酸)显著短于外周亚型 CRFR2B(54aa),在中枢神经系统呈现高密度特异性分布,主 要富集于下丘脑室旁核(PVN)、杏仁核中央核(CeA)、海马 CA3 区及中缝背核。作为 应激恢复的核心介质,该受体被内源性配体 尿皮质素 3(UCN3)高选择性激活,通过 Gs- cAMP-PKA 信号轴 发挥核心功能。 | |
