I. Background
整合素αvβ6 是由αv 和β6 亚基组成的异二聚体跨膜受体，属于整合素家族成员，其胞外域通过识别含 RGD（精氨酸-甘氨酸-天冬氨酸）序列的配体介导细胞与细胞外基质的相互作用，主要在上皮细胞中低表达，但在组织损伤、纤维化、炎症及恶性肿瘤中显著上调，成为疾病治疗的潜在靶点。该整合素的主要配体包括细胞外基质蛋白如纤维连接蛋白（Fibronectin）、层粘连蛋白（Laminin）以及潜活形式的 TGF-β，后者通过与αvβ6 结合被激活为活性形式，驱动纤维化和免疫抑制微环境形成；此外，αvβ6 还可作为口蹄疫病毒（FMDV）等病原体的入侵受体。 αvβ6 通过激活 TGF-β/Smad 通路促进成纤维细胞增殖和胶原沉积，参与特发性肺纤维化（IPF）和肝纤维化的发展；同时，其介导的 FAK/Src 和 MAPK 通路激活可增强肿瘤细胞迁移、侵袭及上皮-间质转化（EMT），在胰腺癌、结肠癌及头颈鳞癌中与转移和不良预后密切相关。此外，αvβ6 还能通过 PI3K/AKT 通路抑制凋亡并促进肿瘤细胞存活，并通过 TGF-β依赖性机制抑制 T 细胞功能，促进肿瘤免疫逃逸。αvβ6 的高表达与多种病理过程相关，包括纤维化疾病（如 IPF、肝纤维化）、恶性肿瘤（如胰腺导管腺癌、三阴性乳腺癌）以及炎症性疾病（如炎症性肠病和类风湿性关节炎）。

II. Description
