| 肿瘤坏死因子-α(TNF-α)是一种分子量为26 kDa 的跨膜蛋白,其前肽在TNF-α-转换酶(TACE)裂解时以可溶性17 kDa 分子的形式分泌。虽然活化的巨噬细胞是TNF-α的主 要来源,但它也可以由多种其他细胞产生,如成纤维细胞、星形胶质细胞、平滑肌细胞、 角化细胞和多种肿瘤细胞,包括B 细胞淋巴瘤、乳腺癌和结肠癌。 TNF-α作为肿瘤促进因子,参与肿瘤发生的所有步骤,包括肿瘤的转化、增殖、血管生成、侵袭和转移。TNF α信号传导也是免疫系统的重要组成部分,可抑制肿瘤发生,阻止病毒复制,是一种内源性热原,可诱导发烧和细胞凋亡。 |
