| I. Background | |
| IL-17 家族是一个包含 6 种细胞因子的家族，包括 IL-17A、IL-17B、IL-17C、IL-17D、 IL-17E（也称 IL-25）和 IL-17F。其中 IL-17A 是促炎信号的关键驱动因子，由辅助性 T17 （Th17）细胞分泌，在自身免疫疾病中起着重要作用。该家族细胞因子在免疫反应和炎症 中发挥作用，并可能通过激活多种通路来介导信号转导。IL17 受体家族包含 5 个成员（IL- 17RA 至 RE），其中 IL-17RA 是通用亚基，与配体特异性亚基（IL-17RB 结合 IL-25、IL- 17RC 结合 IL-17A/F、IL-17RE 结合 IL-17C）形成异源二聚体复合物。 配体结合后，受体通过胞内保守的 SEFIR 基序招募衔接蛋白 ACT1，激活 TRAF6/TAK1 依赖性信号级联，驱动 NF-κB 和 MAPK 通路活化，诱导促炎因子、抗菌肽 及基质金属蛋白酶（MMPs）表达。其过度激活直接导致银屑病、哮喘等病理过程。 |
