| I. Background | |
| TPOR即血小板生成素受体（Thrombopoietin Receptor），由MPL原癌基因编码，也称为CD110、c-Mpl或MPL。它属于I型细胞因子受体家族，是调控造血干细胞自我更新、巨核细胞分化和血小板生成的核心受体。其配体TPO结合后，TPOR发生同源二聚化并激活下游JAK2激酶，进而启动STAT、PI3K等信号通路。该受体约635个氨基酸，分子量约71 kDa，广泛表达于造血干细胞、巨核细胞及血小板表面。在临床上，TPOR是重要的药物靶点，相关激动剂（如艾曲泊帕、罗米司亭）已用于治疗血小板减少症，而其基因突变则与骨髓增殖性肿瘤及家族性血小板增多症相关。 | |
